Characterization of ¥â - Galactosidase from ¢¥ Fuji ¢¥ Apple Fruits



Abstract
¥â-Galactosidase was separated from ¢¥Fuji¢¥ apple fruits by gel filtration, ion exchange column chromatography and characterized by means of several biochemical methods. The results obtained are summarized as follows:
1. One form of ¥â-galactosidase was detected.
2. The Km and Vmax values of ¥â-galactosidase were 3.5 ¡¿ 10^(-2) mM and 3.6 ¡¿ 10^(-2) mmole ¥ñ-nitrophenyl-¥â-D-galactopyranoside/§¢/15 min, respectively.
3. The ¥â-galactosidase was stable below 45¡É with the highest activity at 35¡É.
4. The ¥â-galactosidase was stable at pH between 3 and 7 with the highest activity at pH between 4 and 5.
5. The ¥â-galactosidase activity was inhibited by Ag^+, SDS, or EDTA.
